About Eloralintide
Structure and research context
Eloralintide (LY3841136) is a synthetic, long-acting amylin analogue designed as a selective agonist of the amylin 1 receptor (AMY1R), one of the heterodimeric amylin and calcitonin receptor complexes. It is one of a second generation of non-aggregating, long-acting amylin analogues whose design drew on structural understanding of these receptor complexes and on amylin structure-function relationships. The peptide has the formula C201H319N49O65S2 and a molar mass of about 4,526 g/mol.
Eloralintide in the research literature
In recombinant cell lines expressing individual amylin or calcitonin receptors, eloralintide preferentially activated AMY1R, at roughly 12-fold over the calcitonin receptor (CTR) and 11-fold over AMY3R, whereas at the rat receptors both AMY1R and AMY3R were activated more potently than CTR. This receptor profile distinguishes it from non-selective amylin and calcitonin receptor agonists such as cagrilintide.
It has also been studied in rodent models of energy balance, with pharmacokinetic profiling carried out in rats and monkeys.
