About SELANK / SEMAX Blend
Structure and research context
This blend combines two synthetic heptapeptides, both classed as glyprolines, a group of regulatory peptides that share a C-terminal Pro-Gly-Pro motif. Semax (Met-Glu-His-Phe-Pro-Gly-Pro) is an analog of ACTH(4-10) in which the ACTH(4-7) fragment is extended by Pro-Gly-Pro. Selank (Thr-Lys-Pro-Arg-Pro-Gly-Pro) is an analog of tuftsin, an endogenous immunomodulatory peptide, built on the same tripeptide tail. The two components differ in their N-terminal segments and share the glyproline C-terminus.
SELANK / SEMAX Blend in the research literature
Several studies have examined the two peptides together. In serum enzyme assays, semax and selank both inhibited enkephalin-degrading enzymes in a concentration-dependent manner, with IC50 values of about 10 and 20 micromolar respectively. Their pentapeptide fragments also showed inhibitory activity, while the tri-, tetra- and hexapeptide fragments did not.
In a rat 6-hydroxydopamine lesion model of dopaminergic neuron damage, neither peptide changed motor activity on the elevated plus maze. Tracer work with tritium-labeled peptides mapped how each is broken down. Selank in blood plasma yielded mainly Thr-Lys-Pro-Arg-Pro, Thr-Lys-Pro, Arg-Pro and Gly-Pro, while semax incubated with nerve cells yielded mainly His-Phe-Pro-Gly-Pro and Pro-Gly-Pro.
