CJC-1295 (with DAC)

CJC-1295 with DAC is an analog of the 1-29 fragment of growth hormone-releasing hormone carrying a reactive maleimide group, so that after entering circulation it forms a covalent conjugate with the free thiol at Cys34 of serum albumin. It is a tetrasubstituted form of GRF(1-29) with an added N-epsilon-3-maleimidopropionamide derivative of lysine at the C-terminus. Its molecular weight is about 3,647 g/mol (formula C165H269N47O46, CAS 446262-90-4). The albumin-binding moiety is the feature that distinguishes it from the unconjugated analog.

CJC-1295 (with DAC) in the research literature

The compound was identified from a set of three maleimido derivatives of GRF(1-29) conjugated to serum albumin ex vivo. All three conjugates showed greater in vitro stability against dipeptidylpeptidase-IV and were active in a growth hormone secretion assay on cultured rat anterior pituitary cells. In normal male Sprague Dawley rats the selected compound produced roughly a fourfold larger growth hormone area under the curve over two hours than unmodified GRF(1-29), and immunoreactive material remained detectable in plasma beyond 72 hours; Western blotting showed the compound migrating with the serum albumin band.

In mice with the GHRH gene ablated, repeated exposure over five weeks was associated with growth measures in the normal range and a rise in pituitary total RNA and growth hormone mRNA, consistent with somatotroph proliferation seen on immunohistochemistry.
