Gonadorelin acetate is the synthetic form of gonadotropin-releasing hormone, the decapeptide pGlu-His-Trp-Ser-Tyr-Gly-Leu-Arg-Pro-Gly-NH2, with CAS 103065-82-3, molecular weight about 1,182 g/mol and formula C55H75N17O13. It binds the type I GnRH receptor, a G protein-coupled receptor on pituitary gonadotrope cells. More than twenty naturally occurring variants of the mammalian decapeptide have been identified across species, which may compete for binding at that receptor or engage receptors of their own.
Gonadorelin Acetate in the research literature
Signalling at the type I receptor proceeds through Gq/G11 and the sequential activation of phospholipase C-beta, phospholipase A2 and phospholipase D, generating inositol trisphosphate, diacylglycerol and arachidonic acid; these mobilise calcium and activate protein kinase C isoforms, which in turn drive ERK, JNK and p38 MAP kinase cascades. Early in vitro work found that luteinising hormone release in response to the decapeptide occurred even in calcium-free medium, prompting the proposal that the receptor protein exists in allosteric forms with differing binding activities.
